| 抄録: | 本研究では、非天然アミノ酸導入技術を用いることで、天然および非天然アミノ酸への置換の影響を効率的・系統的に調べることのできる手法の開発や、蛍光標識非天然アミノ酸を導入してタンパク質の構造や機能を蛍光変化や蛍光共鳴エネルギー移動を用いて解析できる手法の開発などを行なった。また得られたタンパク質を診断薬などへ応用し、そのための非天然アミノ酸導入タンパク質の大量合成技術の開発も行なった。 : In this study, an improved method for saturation mutagenesis allowing the substitution with natural and nonnatural amino acids to investigate the effect of the mutagenesis on proteins was developed. In addition, we developed a method for structural and functional protein analysis by introducing fluorescent non-natural amino acids. Practical application of nonnatural amino acid-containing proteins as analytical tools and large-scale production of nonnatural proteins were also investigated. |
